Antipsychotic Drugs and Dysregulated Glucose Homeostasis

Is there an association between antipsychotic drug (AP) use and glucose homeostasis disruptions?In this systematic review and meta-analysis, AP treatment was associated with significant increases in fasting glucose, fasting insulin, and hemoglobin A and elevated hyperglycemia risk compared to placebo in patients with severe mental illness and healthy volunteers. These changes were largely independent of diagnosis; AP type, dose, and treatment duration; age; concomitant medication use; and previous AP exposure.AP-induced dysglycemia puts a population already vulnerable to metabolic dysregulation at even greater risk for long-term health consequences; the current findings should motivate development of new APs with fewer cardiometabolic adverse effects and/or more effective metabolic interventions.
